Transformation of Monascus purpureus to hygromycin B resistance with cosmid pMOcosX reduces fertility

چکیده
∗ Corresponding author Albino strain KB20M1 of Monascus purpureus was genetically transformed to hygromycin B resistance with cosmid pMOcosX, using biolistic bombardment. Conidia and mycelial fragments were used as the recipient material and rupture disk strengths of 6.2 and 9.3 MPa each yielded a transformed isolate from five bombardments. Southern analysis suggested a single copy of the cosmid had integrated into each of the transformants. Both of the independent transformants formed cleistothecia but ascospore formation was greatly reduced or absent, suggesting the integration and expression of the genes carried on pMOcosX interfered with fertilization and/or ascospore formation in this homothallic fungus.

The Effect of Replacing Sodium Nitrite with Monascus Purpureus Pigment in German Sausage
Background: Many common cancers, such as prostate, breast, and colon, are caused by harmful substances and compounds contained in processed foods, including nitrite-treated meats. The aim of this study was to investigate the possibility of replacing sodium nitrite with Monascus purpureus pigment in sausage and to assess its antimicrobial, antioxidant, color, and sensory properties. Methods: The...
متن کاملConstruction of a replacement vector to disrupt pksCT gene for the mycotoxin citrinin biosynthesis in Monascus aurantiacus and maintain food red pigment production.
More and more people pay attention to citrinin produced by Monascus, which has nephrotoxic activity in mammals. It was reported that pksCT gene is responsible for citrinin biosynthesis in Monascus purpureus. In this paper, two DNA fragments in both ends of pksCT were amplified by genomic PCR from fourteen Monascus spp. strains. The PCR products were gained from all of the strains. It is suggest...
متن کاملEfficacy and safety of Monascus purpureus Went rice in subjects with hyperlipidemia.
OBJECTIVE The purpose of this study was to assess the lipid-lowering effect of Monascus purpureus Went rice on serum lipids in patients with hyperlipidemia, and to assess its safety by reporting adverse events and clinical laboratory measurements. DESIGN AND METHODS This was a randomized, double-blind, placebo-controlled study. In all, 79 patients (aged 23-65 years) with a mean baseline low-d...
متن کاملThe Neurospora crassa genome: cosmid libraries sorted by chromosome.
A Neurospora crassa cosmid library of 12,000 clones (at least nine genome equivalents) has been created using an improved cosmid vector pLorist6Xh, which contains a bacteriophage lambda origin of replication for low-copy-number replication in bacteria and the hygromycin phosphotransferase marker for direct selection in fungi. The electrophoretic karyotype of the seven chromosomes comprising the...
متن کاملMonascus purpureus induced apoptosis on gastric cancer cell by scavenging mitochondrial reactive oxygen species
Monascus purpureus is a red dye derived from yeast rice and has been used as color additives for food in East Asia. Monascus purpureus consists of several bioactive components. Some of these components work as a radical scavenger, thus monascus purpureus would also eliminate reactive oxygen species. Cancer cells maintain the high level of reactive oxygen species than normal cell and are death b...
